This video discusses the fundamentals of PCR as it relates to the ABE labs. Colony PCR is an optional extension to the ABE Lab series. PCR allows for the amplification of a target area of a specific genome (for instance rfp).

This video reviews key content for Lab 2/2A: Preparing to Clone a Gene, including the concepts of restriction enzymes and recombinant plasmids. It goes on to explain how these concepts are put into practice during Lab 2 when students use restriction enzymes to begin to engineer a plasmid, as well as several tips and tricks such as minimizing the number of freeze-thaw cycles for your reagents.

This video reviews Lab 4/4a "Verifying the Recombinant Plasmid". In lab 4, students use gel electrophoresis to verify their results from earlier labs, confirming that their ligation was successful and that the recombinant plasmid now includes the genes of interest. This video also provides several tips for conducting this lab with students.

This video contains an overview of ABE's Lab 5/5A: "Transforming Bacteria with the Ligation Products". Once a recombinant plasmid that includes the gene of interest has been created, the next step is to replicate the plasmid and allow the bacteria to produce the protein.

This video provides a detailed overview of ABE's Lab 6: Purifying the Fluorescent Protein. In the first part of this laboratory, students use a reagent called "lysis buffer" to lyse (break open) the cells. In the second part of this laboratory, students use column chromatography to separate out the red fluorescent protein from the sample.
